La flotte comprend deux prototypes XB-52 et YB-52, et 742 appareils de série répartis en huit versions, du B-52A au B-52H. En avril 1952, le deuxième prototype, YB-52, est le premier appareil à voler, suivi à la fin de l'année par le XB-52.
